low-pathogenic. However, this subtype remains a concern for the WHO due to its ability to undergo rapid genetic evolution and active reassortment with highly pathogenic avian influenza viruses. In this regard, there is a clear need to develop effective vaccines against this virus subtype. It is evident that nucleic acid-based vaccines possess numerous significant advantages in comparison to other vaccine types. These vaccines have the capacity to stimulate both humoral and T-cell immune responses and can be rapidly developed; their large-scale production is relatively straightforward and versatile, requiring neither work with dangerous pathogens nor high economic costs. <bold>The aim</bold> of this study was to develop a DNA vaccine against the disease caused by avian influenza virus subtype A/H9 and to evaluate its immunogenicity in a BALB/c mouse model. <bold>Materials and methods.</bold> In the present study, an immunogen encoding the hemagglutinin of the avian influenza virus subtype A/H9 was designed and produced. The transmembrane and cytoplasmic domains were excised from the native sequence, and a number of mutations were introduced to stabilize the protein. In order to obtain an immunogen structure corresponding to the native one, a T4 bacteriophage trimerization domain was added to the C-terminus. The H9 gene, which is responsible for encoding the developed immunogen, was then cloned into the pVAX1 vector. The resulting construct was named pVAX-H9. <bold>Results.</bold> Analysis of the immunogenicity of the construct demonstrated that the vaccine has the capacity to stimulate both humoral and T-cell immune responses when administered to BALB/c mice at a dose of 100 μg. Concurrently, the number of splenocytes producing IFN-γ in response to specific stimulation in the group immunized with pVAX-H9 was twice as high as in the group immunized with the recombinant protein. <bold>Conclusions.</bold> The results obtained in this study indicate the potential of the developed DNA vaccine and of the approach used for immunogen design. The pVAX-H9 DNA vaccine can be regarded as both a single vaccine and a component of combined immunization regimens.</p></abstract><trans-abstract xml:lang="ru"><p>Подтип вируса гриппа птиц A/H9 отличается низкой патогенностью, но, несмотря на это, вызывает обеспокоенность ВОЗ из-за своей способности к быстрой генетической эволюции и активной реассортации с высокопатогенными вирусами гриппа птиц. Установлено, что вирусы А/H9 стали донорами сегментов генов для нескольких вызывающих опасения вирусов, включая А/H5N1 и А/H7N9. Нельзя исключать и потенциальное увеличение вирулентности вируса гриппа птиц подтипа А/H9 за счёт приобретения мутаций, влияющих на репликацию вируса, его стабильность, патогенность и интенсивность внутривидовой и межвидовой передачи. В связи с этим очевидна необходимость разработки эффективных вакцин против данного подтипа вируса. Вакцины на основе нуклеиновых кислот обладают рядом весьма важных преимуществ по сравнению с другими типами вакцин. Они способны стимулировать как гуморальный, так и Т-клеточный иммунный ответ, могут быть быстро наработаны, их крупномасштабное производство относительно простое и носит универсальный характер, не требуя работы с опасным патогеном и высоких экономических затрат. ДНК-вакцины – это платформенная технология, которая даёт возможность быстро менять целевой ген в конструкции без изменения условий производства. <bold>Целью</bold> данной работы были разработка ДНК-вакцины против заболевания, вызываемого ВГП подтипа А/Н9, и анализ её иммуногенности на модели мышей линии BALB/c. <bold>Материалы и методы.</bold> В нашей работе был проведен дизайн и получение иммуногена, кодирующего гемагглютинин вируса гриппа птиц подтипа А/Н9. Из нативной последовательности были удалены трансмембранный и цитоплазматический домены, а также внесен ряд мутаций для стабилизации белка. Для получения структуры иммуногена, соответствующей нативной, на С-конец был добавлен тримеризующий домен бактериофага Т4. Ген <italic>h</italic><italic>9</italic>, кодирующий разработанный иммуноген, был клонирован в составе вектора pVAX1, и полученная конструкция была названа pVAX-H9. <bold>Результаты.</bold> Анализ иммуногенности полученной конструкции показал, что вакцина способна стимулировать как гуморальный, так и Т-клеточный иммунный ответ при иммунизации мышей линии BALB/c в дозе 100 мкг. При этом, количество спленоцитов, продуцирующих IFN-γ в ответ на специфическую стимуляцию, в группе, иммунизированной pVAX-H9, было в 2 раза выше, чем в группе, иммунизированной рекомбинантным белком.
