IMMUNE RESPONSES TO HERPESVIRUSES AND MULTIPLE SCLEROSIS

Abstract

The review presents publications mainly for the last 5–7 years, devoted to the further study of the role of herpesviruses in the pathogenesis of multiple sclerosis. Information on the features of the immune response to the Epstein-Barr virus and human herpes virus type 6 with multiple sclerosis is discussed. Hypotheses concerning the involvement of these herpesviruses in immunopathological processes in multiple sclerosis are described.

SYSTEMIC LUPUS ERYTHEMATOSUS: ANALYSIS OF POSSIBLE LOCALIZATION OF ACTIVE SITES IN A PROTEIN SEQUENCE OF MONOCLONAL LIGHT CHAIN (NGTA1-Me-pro) WITH METALLOPROTEASE ACTIVITIES

Abstract

It was shown previously that monoclonal light chains (MLCh) corresponding to the phagemid library of recombinant light chains of immunoglobulin of peripheral blood lymphocytes of patients with systemic lupus erythematosus (SLE) specifically hydrolyze only the myelin basic protein (MBP). The preparations of one of the light chains (NGTA1-Me-pro) demonstrated two optimal pH values, two optimal concentrations of metal ions and two Km values for MBP. Two protease active centers of NGTA1-Me-pro were metal-dependent. In this paper, the homology of protein sequence NGTA1-Me-pro with those for several classical Zn2+- и Ca2+-dependent ap wtll as serine human proteases was analyzed for the first time. The analysis revealed possible protein sequences NGTA1-Me-pro responsible for the binding of MBP, chelation of metal ions and direct catalysis. The data obtained are generalized by means of hypothetical models of the structure of two active centers of the light chain of antibodies.

ASSOCIATIONS OF THE SPECIFIC IMMUNE REACTIONS ON THE CHEMICAL CARCINOGENS AND STEROID HORMONES WITH THE GENE POLYMORPHISMS OF DNA-REPAIR ENZYMES IN BREAST CANCER PATIENTS

Abstract

The supposed interrelations of antibodies specific to low-weight xeno- and endobiotics with DNA-repair genes polymorphisms were studied in postmenopausal healthy women (HW) and breast cancer patients (BCP). Enzyme-linked immunoassay was used for IgA and IgG to benzo[a]pyrene, estradiol and progesterone detection in 215 HW and 344 BCP. Gene polymorphisms of XPD/ERCC2 (rs13181), ADPRT/PARP1 (rs1136410), HOGG1 (rs1052133), XRCC1 (rs25489) were analyzed by allele-specific PCR. No associations of studied gene polymorphisms with breast cancer were revealed, as well as interrelations between immunological and genetic parameters in HW. It was detected that levels of IgG specific to benzo[a]pyrene, estradiol and progesterone were associated with the C allele of ADPRT/PARP1 (p=0.02; p=0.003; p=0.006 accordingly). Therefore, it was confirmed that genes of DNA-repair enzymes could determine formation of antibodies specific to the chemical carcinogens and endogenous steroids.

CHANGES IN THE INTERFERONAL STATUS INDICATORS IN PATIENTS OF THE LOCAL-DISTRIBUTED KIDNEY CANCER IN THE DYNAMICS OF THE DISEASE

Abstract

The aim of the research was to study the indicators of serum α- (IFN-α) and γ-interferon (IFN-γ), the levels of spontaneous and stimulated production of interferons by peripheral blood mononuclear cells in patients with locally advanced renal cell carcinoma (RCC) before and 14 days after the operation. Patients with RCC (T3 N0 M0) were examined before surgical treatment and 14 days after radical nephrectomy. The concentration of IFN-α and IFN-γ in serum was determined using enzyme-linked immunosorbent assay (ELISA). The level of spontaneous and stimulated (concanavalin A) production of IFN-α and IFN-γ was determined in the supernatants of peripheral blood mononuclear cells cultured for 72 hours by ELISA. It was found that in patients with locally advanced RCC, both before surgery and after surgical treatment, an increase in serum levels of IFN-α and IFN-γ was observed, and the spontaneous production of IFN-γ by peripheral blood mononuclear cells was also increased. 14 days after surgery there was a decrease in the spontaneous production of IFN-α in combination with an increased ability of peripheral blood mononuclear cells to induce the production of IFN-α and IFN-γ. The identified imbalance in the IFN-producing system in patients with RCC in the postoperative period must be considered when developing immunorehabilitation programs in this category of patients.

IMPAIRMENT OF THE DIFFERENTIATION OF PERIPHERAL BLOOD AND PERITONEAL FLUID B-LYMPHOCYTES IN PATIENTS WITH ENDOMETRIOSIS AND INFERTILITY

Abstract

It was supposed that endometriosis development is associated with autoimmune disorders, but the character of B-lymphocytes functioning during this gynecological disease practically is not studied. The aim of our work was to elucidate the character of the peripheral and peritoneal B-cells differentiation in patients with endometriosis and infertility. 59 women, who underwent surgical treatment of infertility in gynecological clinic, were carried out into investigation. I–II stages of endometriosis were diagnosed in 35 women and III–IV stages – in 24 women. 31 women, who underwent surgical sterilization, formed a control group. Peripheral blood and peritoneal fluid were used as the material for investigation. Peripheral and peritoneal B-lymphocytes phenotype was estimated by multicolor flow cytometry. It was found that during endometriosis at the systemic and local level the content of plasma cells, unswitched CD19+IgD+CD27– naïve B-lymphocytes was increased and the level of switched CD19+IgDCD27+ memory B-cells was diminished. In peritoneal fluid of women with endometriosis the amount of unswitched CD19+IgD+CD27+ memory B-cells was also decreased. In healthy women the population of peritoneal B-lymphocytes differed from peripheral B-lymphocytes by higher content of unswitched CD19+IgD+CD27+ memory B-cells and lower level of naïve B-lymphocytes. In women with endometriosis at the local level the amount of unswitched CD19+IgD+CD27 naïve B-lymphocytes and switched CD19+IgDCD27+ memory B-cells was lower and levels of CD27IgD and CD27+IgD+ memory Bcells were higher than that in the blood. Likely, the noted elevation of autoantibodies production during endometriosis may be due to increased levels of peripheral and peritoneal naïve B-lymphocytes and diminished amount of peritoneal unswitched memory B-cells, participated in the regulation of the autoantobodies production.

DEPENDENCE OF THE BLOOD T-LYMPHOCYTES PHENOTYPE FROM THE OUTCOME OF THE WIDESPREAD PURULENT PERITONITIS

Abstract

The purpose of the research was to study the phenotypic composition of peripheral blood T-lymphocytes in humans in the dynamics of postoperative treatment depending on the outcome of the widespread purulent peritonitis (WPP). 38 patients with acute surgical diseases and abdominal cavity injuries complicated by WPP were examined. Blood sampling was performed before the operation (preoperative period) and also on the 7th, 14th and 21st day of the postoperative period. The study of the phenotype of blood T-lymphocytes was carried out by flow cytometry. It was established that the phenotype of blood T-lymphocytes in patients with WPP in the pre- and post-operative period differs significantly in depending on the outcome of the disease. With a favorable outcome of WPP in the pre-operative period, an increased number of common T-lymphocytes, including expression of CD62L, and CD4+-T cells, primarily due to Treg (including activated cells) are detected. Consequently, an increase in the amount of Treg in patients with WPP at the peak of the disease is a prognostic sign of a favorable outcome. Treg regulate the intensity of the inflammatory process that leads to an improvement in the clinical state of patients with WPP and accordingly increases the likelihood of a favorable outcome of the disease. The phenotype of blood T-lymphocytes significantly changes in the dynamics of post-operative treatment. Regardless of the outcome of the disease, the content of T-cells expressing CD28 and CD62L, as well as the T-helper and Treg level, is significantly reduced in patients. Only with a favorable outcome of the disease at the end of the observed period (21 days of treatment) in the blood increases the amount of cytotoxic T-lymphocytes and T-cells with CD57 expression. These changes in the phenotypic composition of T-lymphocytes are determined by the use of antibiotics and the migration of cells into the inflammation zone.

NEUTROPHIL FUNCTION IN PATIENTS WITH POSTSURGERY INFECTIOUS SEPTIC COMPLICATIONS

Abstract

Dysfunction of the neutrophils is associated with various infectious processes, particularly, with sepsis and other postsurgical bacterial complications. Quantitative evaluation of the neutrophil function is a promising approach for early diagnosis of infectious and septic complications in clinical practice. The aim of this study was to assess the relationship between functional parameters of neutrophils (CD64 expression and oxidative metabolism) and standard clinical and laboratory markers of inflammation in patients who underwent large intestine resections. We assessed 49 patients with III – IV stage colorectal cancers; we evaluated lactate and C-reactive protein levels, assessed sepsis-related organ failure assessment (SOFA) score and performed bacteriological investigation. CD64 expression was determined by means of flow cytometry; oxidative metabolism was studied by means of chemiluminescent (CL) analysis with double-step activation followed by automatic signal deconvolution. We revealed association between complications and the increase of CD64 expression, as well as increase of luminol-dependent intracellular CL light sum and decrease of the lucigenin-dependent intracellular CL intensity. The progression of sepsis-associated multiple organ failure is also associated with increase of CD64 expression; strong significant correlation between the lactate level and CD64 expression has been shown. The prognostic index of luminol-dependent CL significantly increases only in critical organ dysfunction, while CD64 is more sensitive to multi-organ failure progression.

EXPERIENCE OF INTRA-LABORATORY QUALITY CONTROL APPLICATION AT DETERMINATION OF CLASS G ANTIBODIES TO MEASLES VIRUS BY IMMUNE-ENZYME ANALYSIS METHOD USING CONTROL DRUG

Abstract

Measles is a highly contagious infectious disease, fraught with serious complications and even death. The measles elimination program adopted by WHO has produced tangible benefits, but measles is still not defeated. At the stage of measles elimination, measures to increase the reliability of the results and quality of laboratory tests of antibodies to the measles virus become particularly relevant. The purpose of these studies was the validation of the drug “ILC measles-IgG” for the determination of measles antibodies of class G in serum by ELISA and the development of accounting parameters for qualitative and quantitative research options. The study material was commercial reagent kits for determining the concentration of antibodies to the measles virus VeсtoMeasles-IgG (JSC Vector-Best, Novosibirsk, Russia), which determined the concentration of the desired analyte in blood serums of 654 randomly selected conditionally healthy residents of Moscow and the Moscow region in age from 0 to 60 years. When conducting laboratory quality control, “ILC measles-IgG” (lyophilized preparation containing IgG to measles virus) was used to assess the intra- and inter-series convergence of the analysis. It is shown that certified on the basis of the reference laboratory of WHO / Europe of G. N. Gabrichevsky Research Institute for Epidemiology and Microbiology sample “ILC-measles IgG” is suitable for conducting internal laboratory quality control in determining measles IgG in serum using the “VectoMeasles-IgG” kits by the qualitative (OU) and quantitative (IU / ml) method. When assessing the convergence and reproducibility of the results of “ILC IgG-measles”, the CV values should not exceed 8% and 19%, respectively, and the OD values (OU) and IU / ml of “ILC measles-IgG” working solutions (native or diluted 1: 2) should not be in the range of values (points) of test calibration samples.

BLOOD LEUKOCYTE PHAGOCYTIC AND CYTOKINE-PRODUCING ACTIVITY OF ANTI-PLAGUE VACCINATED BALB/C LINE MICE AGAINST THE BACKGROUND OF IMMUNOMODULATION BY POLYOXIDONIUM

Abstract

Polyoxidonium increases the immunogenicity and protection of the live plague vaccine, but the cellular mechanisms underlying its immunomodulating effect are not well understood. In present study, the blood granulocyte phagocytic activity (PA) of anti-plague vaccinated and not vaccinated BALB / c mice in relation to Yersinia pestis, Escherichia coli and Staphylococcus aureus was investigated using flow cytometry. The results of the phagocytic reaction were taken into account in the microvolumes of whole blood on days 3, 7 and 21 of immunogenesis. Spontaneous and CoA-induced cytokine production (IFN-γ and IL-10) was evaluated in the blood by an ELISA method. In one of the groups of animals, anti-plague vaccination was carried out against the background of immunomodulation with a polyoxidonium. It was established that in intact mice the blood granulocyte PA in relation to Y. pestis cells is twice as low as in experiments with E. coli and S. aureus cells. Anti-plague vaccination specifically activated in vitro the blood granulocyte absorptive capacity only in relation to plague microbes. Against the background of the polyoxidonium immunomodulating effect, PA increased in experiments with Y. pestis cells already on the 3rd day of immunogenesis, which is consistent with the known ability of this immunomodulator to stimulate earlier antibody genesis during anti-plague vaccination. In addition, an increase in phagocytic index values correlated with an increase in spontaneous and induced cytokine production in blood samples of animals vaccinated against the plague. The data obtained reflect the dependence of blood leukocyte PA from the pathogen type, specific antibodies and cytokines. They indicate that the polyoxidonium ability to increase the live plague vaccine protectivity may be associated with the activation of leukocyte PA to plague microbes.

Abstract

T-lymphocytes of vaccinated peopleare involved in the post-vaccination immune response to the polysaccharide vaccine against the A, C, Y and W serogroups of N. meningitidis even if the vaccine is not a glycoprotein conjugate, but the functional activity of T-lymphocytes producing IFNγ and IL-4 is diff erent in the primary and secondary response to the vaccine.If in revaccinated people the humoral immune response to the A-polysaccharide of N. meningitidis is poorly developed, there is the lack of activity of T-lymphocytes that produce IFNγ in their blood and the change in the balance between the activity of Th1 and Th2 is more similar to the dynamics of the primary vaccinated people, rather than successfully revaccinated.

Abstract

The aim of the study was the selection on the basis of clinical and immunological studies of the types of immune system response in patients with bedsores.67 patients with acute surgical diseases complicated by bedsores were examined.The study of the phenotype of blood lymphocytes was performed by flow cytometry.For typing of patients with bedsores the cluster analysis method was used.An immunological examination of patients with bedsores found that immunological indicators are characterized by a significant variety.The use of cluster analysis allowed us to isolate immunotype 4 in patients with bedsoreswhich are defined as different pathogenetic variants of the course of the disease.

VITAMIN D STATUS OF CHILDREN WITH RECURRENT INFECTIONS AND ASTHMA

Abstract

The level of 25(OH) hydroxycholecalciferol in healthy children with recurrent respiratory diseases and patients with mild bronchial asthma was studied. It was found that in children with repeated respiratory infections there was a significant decrease in the concentration of vitamin D against the background of an increase in total serum immunoglobulin E in comparison with a group of healthy individuals. Patients with bronchial asthma of mild severity demonstrate lower values of vitamin D.

HISTAMINLIBERATION AND CD45 DYNAMIC IN DRUG HYPERSENSITIVITY (DHS) PATIENTS

Abstract

The leucocyte specific activations on the dynamic expression of CD45-antigen (CD45-test) by the flow cytometry, the leucocyte histaminliberation nonspecific and specific (to the local anaesthetics (LA)) in the DHS patients were assessed in vitro. The leucocyte activation in the DHS patients was revealed in 80% by CD45- test. Increased spontaneous leucocyte histaminliberation – in 36,4%, nonspecific stimulated – 67,7%, LA-stimulated – 27,3% pts. КСCD45 were correlated with spontaneous histaminliberation (r = – 0,46, р=0,002), stimulated histamineliberation (r = – 0,43, р=0,013) и LA-stimulated histamineliberation (r = – 0,28, p = 0,07).

CHARACTERISTIC OF SENSIBILISATION SPECTRUM IN ATOPIC

Abstract

The spIgE to the different allergens and CD45 antigen dynamic (KCD45) to the drug in vitro were assessed in atopic. The increased spIgE levels to the plant (30,8%), epidermal (24,6%) and food (25,5%) allergens were revealed. The CD45-test with the anaеsthetics were most popular (76%), among the local anaеsthetics were 61,8% cases. The increased value KCD45 to the lidocain (30%) and ultracaine (21,4%) among local aneasthetics and to the ketamine (20%) and diazepam (17%) among preoperative screening drug were revealed.

We have studied the tissue-specific features of the expression of the NAMPT gene encoding visfatin. In patients with obesity with type 2 diabetes, a violation of the insulin-protective mechanism of the effect of visfatin on tissue is recorded, which is confirmed by the positive correlations of the level of visfatin with leptin and insulin in the circulation.

MECHANISMS OF FORMATION OF BRONCHIAL OBSTRUCTION IN CHILDREN OF PRESСHOOL AGE

Abstract

Investigated the indices of some cytokines and chemokines in peripheral blood serum (IL-1β, IL-6, IL-8, IL-10, IFN-α, IFN-β, IFN-γ, TFN-α, MCP-1, RANTES, MIP-1α, MIP-1β) in children with acute bronchitis. When evaluating concentrations of cytokines in peripheral blood serum of the patients with acute bronchitis in general (n = 87 people) we revealed significantly higher levels of IL-8, IFN-β, MCP-1, RANTES, MIP-1α, MIP-1β, as compared to control group. It was established that MIP-1α и MIP-1β concentration in the group of children with acute simple bronchitis is twice as high as in children with acute obstructive bronchitis. Immunological markers of acute obstructive bronchitis are lower serum concentrations of MIP-1α and MIP-1β, with high IL-10 values, which confirms the significance of the Th2 pathway in the pathogenesis of bronchial obstruction.

GM-CSF-INDUCED GRANULOCYTIC MYELOID REGULATORY CELLS ARE ACTIVATED BY BACTERIAL LIPOPOLYSACCHARIDE TO SUPPRESS HUMAN T-CELL RESPONSES

Abstract

Myeloid-derived suppressor cells (MDSC), initially found in malignant diseases, contributing to the development and / or course of cancer, autoimmune disorders, generalization of infections up to the development of sepsis, are functionally and phenotypically heterogeneous populations – monocytic and granulocytic MDSC. Granulocyte MDSC, or granulocytic regulatory cells (Gregs) induced by GMCSF, has been shown to inhibit the proliferative response of allogeneic lymphocytes in the presence of lipopolysaccharide (LPS) or fl agellin in various ways. Thus, in a situation where LPS, but not fl agellin, was present in cultures of allogeneic mixed lymphocytes, the introduction of Gregs induced by GMCSF significantly inhibited the proliferative responses of allogeneic lymphocytes. To the contrary, in the presence of fl agellin, Gregs rather increased the proliferation of responding responder cells, whereas in the absence of Gregs neither LPS nor fl agellin aff ected the proliferative response of responder cells. Determination of the gene expression profile in Gregs induced by GM-CSF or LPS revealed significantly (7–10-fold) increased transcription of genes encoding PTEN and ARGI (P <0.05), but not IL1B or TNF-alpha. It is assumed that microorganisms can use their own LPS in order to ensure PTEN-controlled energetic safety of immunosuppressive Gregs and enhance specific activity of the latter mediated by an ARGI product, the secreted arginase enzyme that limits the availability of arginine for the immune cells. Such a mechanism of immune escape allows the bacterial populations to survive better due to the weakening of antibacterial immunity.

Abstract

Persistence of high-risk human papillomavirus (HPV) genotypes is the leading cause of cervical cancer, as well as anogenital and oropharyngeal cancers. This article focuses on analysis of the global use of the licensed HPV vaccines, given the introduction of HPV vaccination into national immunization programs of many countries. All vaccines are safe, immunogenic and effective in population usage, with the most frequent adverse event being injection-site reactions.

Abstract

Persistence of high-risk human papillomavirus (HPV) genotypes is the leading cause of cervical cancer, as well as anogenital and oropharyngeal cancers. This article focuses on analysis of the global use of the licensed HPV vaccines, given the introduction of HPV vaccination into national immunization programs of many countries. All vaccines are safe, immunogenic and effective in population usage, with the most frequent adverse event being injection-site reactions.

Abstract

Aims: to identify phenotypes of the bronchial asthma (BA) upon admission of patients for the sanatorium medical rehabilitation (SMR) using cluster analysis and to analyse their pathogenetic features. The research include 518 patients with BA who passed SMR. Survey conducted: clinical, functional, laboratory. Immunological indicators were studied: IGE, IFN-α, IFN-γ, IL-4, EGF, TGF-β, TNF. Statistical processing of material in the SPSS25 program. On the basis of results of inspection of patients with cluster analysis is BA applied and 3 clusters are selected. Diagnostic parameters for reference of patients to clusters are determined and features of immunological inspection are described. Selection of homogeneous groups at a stage of rehabilitation matters for development of the personalized programs of rehabilitation.

Abstract

In work secretion by neutrophils the preforming of cytokins after an hour incubation to the RPMI 1640 environment is studied. Secretion of cytokin was defined with use of multiplex sets of the BioRad company (USA) for definition of 17 cytokin (G-CSF, GM-CSF, IL-10, IL-12p70, INF-γ, IL-13, IL-17A, IL-1β, IL-2, IL-4, IL-5, IL-6, IL-7, TNF-α, IL-8, MCP-1, MIP-1β) on the MAGPIX-100 device (USA). It is established that at an incubation of neutrophils in the environment of RPMI 1640 within 1 hour in the environment of an incubation (supernatant) come concentration of which in a supenatant of neutrophils exceeded GM-CSF, IL-12p70, IL-17A, IL-1β, IL-6 IL-8 and MIP-1β reference level (RPMI-1640 environment) by 1.2–7.0 times (p <0.05–0.001). Thus, neutrophils are able to secrete preformed cytokines into the culture medium for 1 hour after the start of their incubation.

Abstract

In a research approaches to assessment of levels of cytokin of neutrophils on the basis of secretion by neutrophils the preforming of cytokins after an hour incubation are offered the RPMI 1640 environment. Secretion of cytokins was defined with use of multiplex sets of the Biorad company (USA), Luminex (USA), for definition of various cytokins on the MAGPIX-100 device (USA) and the immunofermental analysis (IFA). It is established that at an incubation of neutrophils in the environment of RPMI 1640 within 1 hour in the environment of an incubation (supernatant) various cytokins come to light concentration of which in a supenatant of neutrophils exceeded reference level (RPMI-1640 environment). At addition to neutrophils of various bacteria, supernatant of bacteria, regulatory molecules, the activity of secretion changes (is stimulated or supressed) depending on that substrate which was added to the environment of an incubation of live neutrophils. Thus, the offered methodical approach of assessment of levels of cytokin of neutrophils allows to study mechanisms of regulation of cytokins network of these cages.

An extended analysis of the immune status (more than 45 indicators) was carried out in 120 patients with a burn injury. The revealed statistically significant immune disorders are associated with parallel processes: immune hyperactivity and immunosuppression, the severity of which depends on the area of the burn injury and the period of the burn disease. Early immunodiagnostics according to the formula for prediction of sepsis developed by us, allows to identify patients with a high risk of generalization of infection and sepsis for the corresponding complex therapy and timely immunocorrection.

BASOPHIL ACTIVATION TEST IN THE DIAGNOSIS OF CHRONIC URTICARIA

Abstract

Chronic urticaria (CU) is a serious medical and social problem that significantly reduces the quality of patients life. Chronic autoimmune urticaria (CAU) is characterized by the presence of autoantibodies to IgE or its high affinity FcεRI receptor. The study included 16 patients with CU. The autologous serum skin test (ASST) was positive in 31% of patients. The level of elevated total IgE was detected in 38% of patients. A higher number of basophils with activation markers CD203c (14.0 ± 2.7% vs 9.3 ± 2.3%) and increased values of the stimulation index (SI) (2.0 ± 0.4 vs 1.3 ± 0, 3) were identified in patients with positive ASST compared to patients with negative ASST. The results of intracutaneous testing and positive correlation with expression of basophil activation markers made it possible to diagnose CAU in 64% of cases. Basophil activation test in vitro is a promising method for the diagnosis of CAU, which allows to determine further therapeutic tactic.

In the study estimation of the informative defination of the activity ratio of mitochondrial dehydrogenases glycerol-3-phosphate (GPDG) and succinate (SDG) in the major populations of lymphocytes to predict graft rejection at the 15 recipients of the group’s risk before rekeratoplasty. An increase in the ratio of GPDG/SDG activity in all major lymphocyte populations, most pronounced in B-lymphocytes and NK- cells, was found in patients with adverse outcome. The ratio of activity GPDG/SDG can serve as a prognostic indicator of graft rejection in keratoplasty.

Abstract

The primary open-angle glaucoma is a first in disability among eye diseases. First-line drugs – prostaglandin analogues are not always effective. The aim of the work was the search for predictors of the ineffectiveness of this therapy. The cytokines TNF-α, TNF-β, IL4, IL-6, IL-10, IL-13, IL-17, IL-22, IL-29, IFN-γ, TGF-α, TGF-β1, TGF -β2 was investigated in the tear fluid of 124 eyes in the diagnosis of glaucoma. Predictors of non-compensation of intraocular pressure when prescribing prostaglandin drugs is a high level of IL-17 and a decrease in TGF-β2. The high level of IL-6 is a predictor of a decrease in visual function during prostaglandin therapy.

DETERMINATION OF THE LEVEL OF CD64 ON NEUTROFILES IN PATIENTS WITH SURGICAL SEPSIS

Abstract

The level of the expression of CD64 on neutrophils by fl ow cytometry in 49 patients with sepsis was studied, the dependence of these parameters on the degree of severity of the condition on the scale APACHEII (in points), the severity of multiple organ dysfunction scale SOFA (in points).High-level of expression on neutrophils nCD64 directly correlated with more severe organ dysfunction (SOFA), the weight of the state (APACHEII). These results may give the suggestion that presence of CD64 on neutrophils may serve as a biomarker refl ecting on the degree of systemic infl ammation in sepsis and may be a criterion in the prediction of the severity of disease and outcome in patients with sepsis.

Abstract

The reactions of mononuclear blood cells (MNC) were evaluated under cocultivation with titanium substrates carrying a bilateral calcium phosphate coating (CP) under conditions of additional activation of the T-cell receptor. It has been established that in the activation 3D model, in contact with CP-coating, the secretion of pro- and anti-inflammatory cytokines (IL-2, TNF-α, IL-10, IL-4) increases, against the background of a decrease in the viability of the MNC culture and changes in their immunophenotype, in the direction of increasing the number of cells expressing CD95+ and CD45R0+ (the number of CD25+ cells has not changed), which may indicate that the MNC is hyperactive in the 3D model when it is stimulated by an activator of T cells.

Abstract

The problem of early postoperative complications after coronary artery bypass surgery remains unresolved, one of these complications is acute kidney injury (AKI), the prevalence of which according to diff erent authors reaches 30%. Purpose: to evaluate the diagnostic and prognostic value in determining the level of matrix metalloproteinase 8 and their type 1 inhibitor (MMP-8, TIMP 1) in patients with acute kidney injury before and after cardiac surgery. Materials and methods: serum of 70 patients with coronary heart disease before and after cardiac surgery divided into 3 groups: with AKI (I gr.), with AKI and diabetes mellitus (II gr.), without renal complications (III gr.). Results: an increase in serum MMP-8 concentration was registered during the development of AKI in I gr. (p <0.05), low MMP-8 level was detected before the operation in group II (p <0.05), an increase in the level of TIMP 1 after surgery was recorded in all groups (p < 0.05).

Abstract

We investigated the direct effect of interleukin-8 (IL-8) on the functional activity of human monocytes/macrophages and T-lymphocytes. It was shown that IL-8enhanced the production of IL-6 and IL-1β by LPS-activated monocytes/macrophages. In addition, IL-8 enhanced the production of IL-2 and suppressed the production of IL-10 and IL-4 by activated T-lymphocytes. The results demonstrate the ability of IL-8 to stimulate both the pro-inflammatory (M1) activity of monocytes/macrophages, and to participate in the direct regulation of adaptive T-cell responses.

The multidirectional content of IL-28 (IFN-lambda 3) and IL-29 (IFN-lambda 1) in serum of venous blood was determined. In HPV, a decrease in IFN-lambda-1 was determined with a statistically significant increase in IFN-lambda-3 in blood serum.

Abstract

The main goal of this work is to highlight the current state of immunomodulatory therapy with targeted – targeted action, which is used effectively when conducting immunomodulatory therapy of secondary immunodeficiencies, provided that the immunomodulatory drug acts on a known receptor. For example, glucosaminylmuramipeptide acts through NOD2 or recombinant interferon alpha 2b with an antioxidant complex through the IFN alpha receptor. Modern data on targeted action drugs – monoclonal antibodies (monAT), currently used are given. The positive and negative effects of biologic therapy are described, as well as methods of preventing and preventing side effects. It has been suggested that in the future, in order to avoid errors and complications, monotherapy should be administered by clinical immunology (in collaboration with specialists from other fields of medicine) under the control of the immune system and interferon status.

Serological testing for the presence of immunity after immunization among vaccinated health workers indicates presence of immunity in 74.7% with a protective antibody titer from 10 to 200 IU / ml in 50.8%.

DYNAMICS OF CYTOKINE LEVELS IN BLOOD SERUM IN PATIENTS WITH METABOLIC SYNDROME USING STATINS

Abstract

The purpose of this research is to study the dynamics of the content in serum of IL-1β, IL-1α, IL-1ra, IL-18, IL-18BP, IL-37, IL-6, sIL-6r, LIF, sLIFr, IGF-1, TNF-α, sTNF -RI, IL-17, IL-2, IL-4, IL-10, TGF-β1, IL-8, CX3CL1, CXCL10, INFγ, M-CSF, IL-34, VEGF-A, EPO in patients with metabolic syndrome (MS) using statins. The use of statins for 6 months in patients with MS is characterized by a decrease in M-CSF to 399 [375–440] pg / ml (p <0.001) with a secondary increase in the concentration of the protective factor – sertuina-1 (p<0.001) and HDL, that blocks progression of atherosclerotic processes. The concentration of M-CSF in the range [395–454] pg / ml cancels its stimulating effect to synthesis of VEGF-A and slows the growth of coronary collaterals that maintaining a high risk of myocardial infarction (this effect is leveled when using statins for more than 6 months). The data obtained confirm the relevance of further research in this direction.

Abstract

In this study, we assessed the blood circulating proinflammatory and inflammatory cytokines and obtained the cut-off level for TNF, IL-22, IL-23, IL-27 between groups of children in exacerbation and remission during treatment with infliximab. There was a significant decrease in serum IFN in children in exacerbation compared with the group of children in remission for both UC and CD. It was found that the younger the age of the child, the faster the antibodies to infliximab were formed. The level of TNFin the blood, the residual level of infl iximab, as well as the level of antibodies to infliximab can serve as predictors of non-response and loss of effect from therapy with TNF blockers.

The study investigated the relationship between the concentration of 25-OH vitamin D, food hypersensitivity and the level of pro-inflammatory cytokines in children with ASD. An inversecorrelation was found between the concentration of vitamin D and specific IgG (sIgG) to food antigens of dairy and grain products, concentration of pro-inflammatory cytokines (IL17, IFNγ). Abstract

Despite the existing immunoreactivity defects in patients with hemoblastosis, the reasons of the relatively low incidence of oropharyngeal candidiasis (OPC) remain unclear. Among the 525 observed patients with hemoblastosis, in 137 of them werenoted clinical signs of OPC. Of these, in 51 patients (9.7%) OPC was verified. The probability of OPC occurrence significantly increased with the use of antibacterial therapy. The OPC development was accompanied by an increase of IL-6, IL-8, SLPI, DEFb2 and a decrease of IL-17 and MCP-1 in the oral fluid, indicating insufficient functional activity of monocytemacrophage cells, as well as regulators of T-cell response in the oral mucosa.

APPLICATION OF IN VITRO CULTURAL MODELS WITH INTERLEUKIN-7 FOR ESTIMATING THE FUNCTIONAL ACTIVITY OF T-LYMPHOCYTES

Abstract

Currently, studies on the action of cytokines on the functional activity of immunocompetent cells are of great scientific interest. The most signifi ant is the use of various models of the immune system cells culture. This paper describes two cellular models used to study the direct effect of interleukin-7 on the functional properties of T-lymphocytes. Interleukin-7 (IL-7) is involved in the main stages of the T-cell life cycle: carrying out the signals necessary for survival, differentiation, proliferative activity, protection from apoptotic death, homeostatic maintenance of the number of T-lymphocytes. The directionality and severity of the effects of IL-7 was evaluated on the proposed homeostatic and activation T-cell models in vitro.

Abstract

The study is devoted to the search criteria for creating new bioresorbable materials that form hematopoietic stem cell niches that provide vascularization of regenerating bone tissue. In the experiment, it was revealed that the formation of hematopoietic stem cells pool under in vitro cultivation conditions depends on the surface roughness index of the artificial matrix.
