FEATURES OF THE FUNCTIONAL STATE OF THE IMMUNE SYSTEM IN PERSONS SUBJECTED TO CHRONIC RADIATION EXPOSURE IN UTERO IN THE REMOTE PERIOD

Abstract

The embryonic and fetal tissues are more radiosensitive than the tissues of an adult organism. Therefore, intrauterine irradiation is a greater danger compared to irradiation in the postnatal period. A complex assessment of the functional state of the immune system was carried out in the period of the long-term effects of chronic radiation exposure in persons irradiated in utero and in early childhood. Various violations of the immune system are shown in persons irradiated in the antenatal period of development, compared with people irradiated in the postnatal period.

THE ROLE OF ADENOSINTRIPOSPHATE IN THE REGULATION OF HUMAN PERIPHERAL BLOOD NEUTROPHILES AND MONOCYTES DEGRANULATION IN VITRO

Abstract

At present, the metabolism of nucleotides is of great interest in the context of the regulation of functions of immunocompetent cells in inflammation. It is adenosine triphosphate that is attributed to many effects that cause chemotaxis, activation, degranulation of various subpopulations of leukocytes. In this study, the effect of ATP on the degranulation of neutrophils and various subpopulations of human peripheral blood monocytes in vitro was studied using flow cytometry. We have shown that the addition of ATP at concentrations of 1 mM and 0.1 mM caused a significant increase in the expression level of the СD66b degranulation marker on neutrophils. An increase in the expression level of the СD63 degranulation marker on a subpopulation of "classical" monocytes expressing СD14 is shown only at the time of 15 min of incubation, while subpopulations of "nonclassical" monocytes expressing СD16 did not respond to the application of this stimulant.

REACTIONS HAVE DIFFERENCES OF PHAGOCYTES OF PERIPHERAL BLOOD OF PATIENTS WITH DIFFERENT VOLUMES AFFECTED LUNG INFILTRATIVE TUBERCULOSIS

Abstract

The purpose of research is the study of differences in the reactions of phagocytes of peripheral blood of patients with different lesion volume infiltrative pulmonary tuberculosis. The work was based on a study of the clinical and laboratory data of 44 patients with infiltrative pulmonary tuberculosis (29 patients were affected lung segment 1-3, 15 patients had tuberculosis 4 and lighter segments). The control group consisted of 25 healthy people. Used method - flow cytometry, Coulter*Epics*XL instrument (Beckman Coulter, USA). Evaluated absorbency peripheral blood phagocytes (monocytes and neutrophils), ability to oxidative exposure, the antigen-presenting and chemotactic functions. Statistical data analysis was performed using the «Microsoft Office Excel 2007" program and «Statistica For Windows v.6.1». It was found that the infiltrative tuberculosis 1-3 segments of the lung accompanied by an increase in the populations of monocytes and granulocytes which express the molecule CD11b adhesion. TB 4 or more segments of the lung is associated with an increase in the number of monocytes and granulocytes expressing CD11c, decrease monocyte production of superoxide anion. Immunological evaluation of lung volume criterion is the percentage of monocytes, producing superoxide anion.

DIFFERENTIATED APPROACH IN EVALUATING THE RESPONSE OF IMMUNE SYSTEM TO THE PRESENCE OF M.TUBERCULOSIS WITH DIFFERENT DRUG SUSCEPTIBILITY

Abstract

The purpose of the study has a comparative study of quantitative and functional state of the immune system cells in patients with infiltrative pulmonary tuberculosis, which is caused by drug-sensitive and drug-resistant disease. The study included 29 patients, of which: from 12 people the disease was caused by drug-sensitive M.tuberculosis, 17 - the causative agent had multidrug resistance and 25 healthy people. We estimated the number of T- (CD45+CD3+) and B-lymphocytes (CD45+CD19+), NK-cells (CD45+CD3CD16+CD56+), T-helper cells (CD3+CD4+), cytotoxic T cells (CD3+CD8+), γδ-T-lymphocytes (CD3brightCD4-), NKT-cells (CD3+CD16+CD56+), activated T-lymphocytes and by the expression of T-helper cells of CD25 and HLA-DR, the probability of apoptosis of CD95 expression, the number of T-reg-cells (CD3+CD4+CD127CD25+) by flow cytometry on Coulter®Epics®XL instrument (Beckman Coulter, USA). Statistical data analysis was performed using Microsoft software, USA (Office Excel 2007) and StatSoft, USA (Statistica For Windows v.6.1.). Established that infiltrative tuberculosis caused by drug-sensitive M.tuberculosis, accompanied by an increase in HLA-DR expression and decrease in CD95 on T-lymphocytes, reducing the amount of B-cells, increased number of NK-cells. Infiltrative tuberculosis caused by M.tuberculosis multidrug resistance, is characterized by the absence of reducing CD95 on T-lymphocytes, a decrease in the population of NKT cells. Immunological criterion for evaluating drug sensitivity M.tuberculosis is the absolute quantity of CD3+HLA-DR+-cells.

ANALYSIS OF THE APOPTOTIC DEATH INDICATORS OF LYMPHOCYTES AMONG THE PERSONS EXPOSED TO CHRONIC IRRADIATION

Abstract

The evaluation of the apoptotic death of lymphocytes among the inhabitants of the coastal villages of the Techa River exposed to long-term low-level irradiation was carried out. We analysed the number of lymphocytes in the stage of DNA fragmentation by the TUNEL method, the number of CD95+ lymphocytes in the blood and the levels of TNFα in serum. A statistically significant increase in serum TNFα was observed in persons exposed to chronic radiation exposure in the dose range from 0.08 to 4.23 Gy.

BLASTIC PLAZMATSITOIDS DENDRITOCELL TUMOUR: EXPERIENCE OF DIAGNOSTICS AND TREATMENT IN THE SVERDLOVSK REGIONAL ONCOHEMATOLOGICAL CENTER

Abstract

Aim: to analyze the case of blastic plasmacytoid dendritic cell neoplasm (BPDN) detection and treatment in Sverdlovsk Regional Hematological Centre (Ekaterinburg). Bone marrow and peripheral blood samples obtained from female, aged 32, treated in Sverdlovsk Regional Hematological Centre during the Jan-Marth 2014. Diagnosis of BPDN verified using cytology, cytochemistry and immunophenotyping. Cytogenetic analysis was realized using G-banding. Detection of mutations in exons 12-13 ASXL1, exons 18-26 DNMT3A, exons 12-15 and 19-21 FLT3, exons 7-12 and 16-19 KIT, exons 1-4 KRAS, exons 9-12 NPM1, exons 1-4 NRAS, exons 4-11 ТР53, exons 6-9 WT1 genes was performed using direct sequencing. Cytological characterization of BPDN blast cells: large cells with oval, reniform nuclei with delicate chromatin structure, with one or two, at least - several small nucleoli, with wide basophilic cytoplasm and bright perinuclear area, richly vacuolated. In cytochemistry lipids is positive in 10.0 % of blasts, glycogen -6.0 %. The primary method of differential diagnosis of BPDN in case study was the identification of blast cells in the bone marrow. Immunophenotype of blast cells was CD4, CD15, CD33, CD38, CD56, CD64, CD65, HLA-DR(+), MPO-cyt (+/-). Karyotype was 48, XX, add(1)(p36), +6, +8 [8]. In case 2 gene mutations were co-existed in samples: KIT gene insertion and TP53 gene non-synonymous substitution с. 215 C>G. Coding sequence the remaining exons of the investigated genes is fully consistent with "wild type".

Abstract

It has been established that, both in females and in males, 6 h of immobilization resulted in a pronounced suppression of the amount of PFC in the spleen. Preliminary blockade of opiate receptors led to the abolition of the effect of stress in both males and females. However, in males, the administration of one naloxone had no effect on the number of PFC, while in females, naloxone inhibited the formation of PFC by both relative and absolute indices.

Abstract

The status of the functional-metabolic status of neutrophilic granulocytes (NG), interleukin (IL) -1β, IL-2, IL-8 in the nasal secretion of patients with 24-hour tamponade at 1,3 days after rhinosurgical intervention for curvature of the nasal septum was analyzed. The nasal secretion of the operated patients revealed: an increase in the content of neutrophilic granulocytes, an increase in the parameters of the neutrophilic granulocyte NGT- test, a decrease in the intensity of the phagocytic reaction intensity, a phagocytic number, an increase in IL-8, IL-2, and a decrease in IL-1-β increased by day 3.

EXPERIMENTAL ESTIMATION OF CORRECTION OF HUMORAL IMMUNITY HEPATOPROTECTIVE DRUG HEPATOBIOL IN LABORATORY ANIMALS C ACUTE TOXIC HEPATITIS

Abstract

The article is devoted to topical issues of preclinical study developed a combined experimental sample of a hepatoprotective drug heptobiol on humoral immunity in laboratory animals and reproduced with a model of acute toxic liver damage. Results showed that the manifestation of the inflammatory syndrome was more pronounced in the group of experimental animals without treatment is reproduced with toxic hepatitis. In group I, in which laboratory animals were assigned to drug heptobiol, the pathological process were less pronounced than in group II received the reference drug ursosan. It should be noted that at the end of the observation period (30th day), group I noted the normalization of all studied parameters, in contrast to group II, in which indicators of inflammatory syndrome were not recovered.

PRECLINICAL EVALUATION OF IMMUNOTROPIC ACTION OF PROBIOTICS BACILACK TRANSDERMAL THERAPEUTIC SYSTEM

Abstract

The article is devoted to the study of immunotropic action developed bicomponent probiotic drug bacilact in a soft medicinal form (based on tizol, eftiderm and silicontitaniumorganical glycerohydrogel) on quantitative changes of parameters of cellular immunity in the treatment of localized infected wound animal skin, caused by Staphylococcus aureus. Thus, in particular, an increase in phagocytic activity of neutrophils of peripheral blood and peritoneal macrophages (average 2 times), increased the quantitative content of T - and B-lymphocytes (1.5-2 times). Studies to assess the immune status of experimental animals with localized purulent infection of the skin caused by Staphylococcus aureus, which treatment the subjects performed gel preparations, allows to make a conclusion about their pronounced immunomodulatory properties on parameters of cellular immunity. More significant effects observed for drugs on the basis of bacilact containing tizol and silicontitaniumorganical glycerohydrogel.

Abstract

The effect of pregnancy-specific glycoprotein (PSG) on the production of Th1/Th2-cytokines by intact mononuclear cells and monocultures of CD4 cells under in vitro conditions by flow-through fluorimetry was studied. It has been established that PSG predominantly suppresses the production of Th1-cytokines (IFN-γ, TNF-α, IL-2) by the types of cells studied, and it regulates the production of Th2-cytokines in different directions, decreasing the production of IL-9, IL-13, IL-6, but increasing the level of IL-5. As a result, PSG forms the integral Th2-cytokine profile of intact peripheral cells of the immune system.

Abstract

Diabetes remains an urgent problem. According to WHO data the number of patients with Diabetum makes 3-5 %. The risk of development of postoperative infectious complications in patients with Diabe-tum is 5 times higher, than at patients without it. The research of hemotaksichesky activity of phagocytes in relation to a glucose and insulin is conducted. The revealed changes of locomotory reactions of phagocytes can demonstrate the blockade of the receptor device of neutrophils and monocytes developing in the conditions of a hyperglycemia.

Abstract

In experiments in vitro on the 23 clinical isolates S. aureus (n=23) a test-system Multiplex (Luminex, USA) to determine the 15 cytokines (G-CSF, GM-CSF, INF-γ, IL-10, IL-12p70, IL-13, IL-17A, IL-β, IL-2, IL-4, IL-5, IL-6, MCP-1, MIP-1β, TGF-α) and test-system studied to determine the 27 cytokines (BasicFGF, Eotaxin, G-CSF, GM-CSF, INF-γ, IL-10, IL-12p70, IL-13, IL-15, IL-17A, IL-1β, IL-1ra, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IP-10, MCP-1, MIP-1a, MIP-β, PDGF-BB, RANTES, TNF-α, VEGF) the production of cytokine-like substances (CLS). Identified and characterized the intergeneric and intraspecific variability of S. aureus in their ability to produce CLS in the environment of cultivation. It is established that isolates of S. aureus are the most active producers of CLS. The possible pathogenetic significance of products CLS S. aureus and ways to further study this phenomenon were discussed.

Abstract

In experiments in vitro and in vivo it was shown that a synthetic peptide of the active site of GM-CSF -ZP2 (chemical formula: THR NLE NLE ALA SER HIS TYR LYS GLN HIS CYS PRO) stimulates the activity of immune system (lymphocytes, stem cells, neutrophils, monocytes), inhibits the growth, reproduction and biofilm formation of gram-positive and gram-negative bacteria and has antiviral activity against DNA and RNA viruses and activates a reparation of wounds. The presence at this synthetic peptide of such a wide range of immunological effects (immunostimulatory, antibacterial, antiviral, and repair activity) determines the possibility of its use as the basis for creation of new effective medicinal and cosmetic preparations with combined properties that was realized at creation of ACEGRAM.

IMMUNOGENETICAL FEATURES AT VIOLATIONS OF REPRODUCTIVE FUNCTION IN WOMEN EXPOSED TO PHENOLS

Abstract

The polymorphic variants of candidate genes associated with immune system, violation of the women reproductive function, as well as with the metabolism of phenols have been analyzed. The study revealed that in the surveyed group, marked by high content of phenols in the blood, the higher frequencies of heterozygous genotypes of VEGF G634C, PPARD A/G, PPARA G2528C, variant homozygous genotypes of TERT C309G and PPARGC1A G1444A genes were detected. It has been noted, that the gene PPARD A/G (p<0.05) characterizes the features in women of this cohort survey. The analysis of the "hapten-genotype" systems, showed an association between the level of phenol, o- and p-cresol, and genotypes of genes: MTHFR C677T, PPARD A/G, ESR1 G2014A. The studied genes can be used as markers of sensitivity that indicates on the preconditions to the development of immunemediated disorders of reproductive function in women induced by an increased content of phenols in blood.

Abstract

To evaluate IgAs, IL-6 and IFN-γ in nasal fluid in transient hypogammaglobulinemia of infancy (THI) 15 children at the age 6 months to 4 years with THI, 30 children with recurrent respiratory infections and 16 healthy persons at the same age were studied. There has been registered a decrease in IgAs and IFN-γ and an increase in IL-6 in THI as compared to healthy children and sufferers of recurrent respiratory infections with no THI. Received data may be interesting for our understanding immunoregulation of antibody synthesis and inflammation in THI.

To evaluate content of IL-4 in lymphoblast transformation test (LTT) supernatants of 15 persons with asthma, 16 patients with asthma in association with allergy to prawns and 15 healthy persons were studied. All sufferers of asthma were sensibilisized to Der p 1 (Dermatophagoides pteronissinus). There have not been revealed any differences between the IL-4 in different groups that may be indirectly propose the presence of Allergen-Associated Molecular Patterns (AAMPs) in patients, which might smooth out received results.

PERIPHERAL BLOOD B CELL SUBSETS IN PATIENTS WITH MULTIPLE SCLEROSIS

Abstract

Demyelination during multiple sclerosis (MS) can be associated with B-cells that can produce au-toreactive antibodies and can be localized in specialized lymphoid follicles within the central nervous system. Using multicolor flow cytometry, it was found out that the relative number of peripheral blood CD19+ B cells in patients with MS (n = 25) was significantly higher (p = 0.005) when compared with the healthy volunteers (n = 27, 17.88 % (12.88, 25.15) and 11.77 % (9.52, 15.26), respectively). To identify the distinct B lymphocytes subsets we studied coexpression of IgD and CD38, IgD and CD27, as well as CD5 and CD27. It was shown that in patients with MS the relative number of IgDdimCD38low "naive" and IgDlow memory subsets was significantly decreased due to the increased proportion of activated IgDdimCD38dim B lymphocytes. Furthermore, the relative content of B cells with the IgDdimCD27low phenotype was significantly increased (p < 0.001), while the percentages of the other subsets purified on the co-expression of IgD and CD27 were significantly reduced. A detailed analysis of B cells and follicular T-helpers - Th subset that affects the maturation and differentiation of B lymphocytes - could have a significant impact on the current understanding of the causes and pathogenesis of MS, as well as could develop the new approaches to the therapy of this disease.

Abstract

The aim of the current study was to investigate cell mechanisms by means of which the "Cytovir-3", the drug for prevention and treatment of the influenza and acute respiratory viral infection, exerts its pharmacological effects. As a cellular model, an endothelial EA.hy926 as well as mononuclear leukocytes (ML) of human peripheral blood were chosen. The effects of co-incubation of "Cytovir-3" and cell lines on IL-8 and ICAM-1 expression were evaluated. In the presence of "Cytovir-3" at concentration of 10.0 ng/ml and 1.0 ng/ml, we found a significant increase in spontaneous secretion of IL-8 by endothelial cells by 10 % and 15 % accordingly. Besides, at concentration of 10.0 ng/ml the investigating drug significantly enhanced the expression level of ICAM-1 induced by TNFa. Revealed stimulating action of "Cytovir-3" on endothelial cells and ML can suggest its influence on vascular component of inflammatory reaction.

Abstract

The aim of the study was to investigate the cell mechanisms mediating immunomodulatory effects of a-glutamyl-tryptophan (EW) and estimate the possibility of this pharmaceutical agent to impact the level of NF-kp phosphorylation. Using in vitro model of inflammation, we tested whether EW impacts spontaneous and TNFa induced IL-8 secretion as well as ICAM-1 expression by endothelial EA.hy 926 cells and mononuclear leukocytes of human peripheral blood. Revealed inhibition of pro-inflammatory cytokine production can contribute to anti-inflammatory effects of EW. However a capability of EW to increase ICAM-1 expression suggests its cell activating function. Thus, due to diverse effects it seems unlikely that EW influences NF-kp directly.

Abstract

Influence of intravitreal injection of vascular endothelial growth factor A inhibitor (VEGF-A) for aged-related macular degeneration treatment on tear fluid immune status, which was evaluated by multiplex analysis with assay of BasicFGF, Eotaxin, G-CSF, GM-CSF, INF-γ, IL-10, IL-12p70, IL-13, IL-15, IL-17A, IL-1β, IL-1ra, IL- 2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IP-10, MCP-1, MIP-1α, MIP-1β, PDGF-BB, RANTES, TNF- a, VEGF was studied. The study showed that given treatment result in a significant decrease in BasicFGF, Eotaxin, IL-13, TNF-α levels in tear fluid in one month.

NANO AND MICRODIMENSIONAL PARTICLES - A NEW LINK IN THE ETIOLOGY OF PERIIMPLANTITA

Abstract

At present, it is being assumed that developments of inflammatory complications accompanying long-term functioning of dental implants in the patient's mouth, are correlated with the development of such diseases as mucositis and peri-implantitis. The pathogenesis of these dental implant complications remains, in our opinion, unexplored in terms of the role of immunological component in the mechanism of acute and chronic inflammation, which in turn requires research aimed at identifying the true causes, which may be proven biologically. It is being observed that, with standardized manufacturing techniques in use when it comes to the practical use of dental implants themselves, the clinical picture of their long-term use is personalized, which in turn can manifest in diminution of bone tissue and require surgical revision or reimplantation. The main causes of these inflammatory complications are correlated with personalized bacterial microenvironment, forming a biofilm on the surface of the dental implant in the patient's mouth, as well as with inadequate distribution of functional pressure connected with the manufacturing and use of the prosthesis. The role of emission of nano and microsized metal particles distributed in the oxide layer of the dental implants manufactured based on the TiO2 alloy and their involvement in long-term interstitial interaction, has not been studied yet. This fact makes it necessary to conduct scientific studies on the immunological aspects of tissue repair at the cellular and molecular levels, and opens up a new perspective on the study of results when it comes to personification of dental implantation.

Abstract

The purpose of this study was to investigate the interaction of nanosized metal particles obtained in the laboratory from the surface of Nobel Biocare dental implants with Pseudomonas aeruginosa, Staphylococcus aureus and Escherichia coli in the presence of native and heat inactivated human serum. In vitro experiments it was established that when nanoparticles were added to test samples of native sera, the accumulated biomass of S. aureus and E. coli was increased by 8.1-8.3 % in broth culture and P aeruginosa by 42.9 % in Comparison of control, indicating that they inhibited the bactericidal effect of the complement system. In addition, the potentiating effect of native serum on biofilm formation of bacteria on the surface of nanoparticles was recorded. Possible immunological consequences of interaction in the system "nanoparticle-whey protein-bacteria in the process of osseointegration" are discussed.

THE INDICES OF IMMUNITY AND THE CLINICAL COURSE OF POSTTRAUMATIC PERIOD IN PATIENTS WITH PENETRATING WOUNDS OF THE EYEBALL WHEN INCLUDED IN A COMPREHENSIVE TREATMENT OF THE DRUG BESTIM

Abstract

We conducted a study of indicators of systemic immunity and the clinical course of post-traumatic period in 38 patients with penetrating wounds of the eyeball severe. In the first group 20 patients received pathogenetic basic treatment, in the second group 18 patients in the standard treatment included intramuscular injection of immunomodulator Bestim 100 mcg daily for 5 days, starting 3 days after the injury. We studied subpopulation composition of lymphocytes in the blood serum on 2-3 and 10 days after injury, the timing of the relief of the inflammatory symptoms, the clinical course of posttraumatic period at 14 and 30 days after injury. It was established that the inclusion of the immunomodulator Bestim in the basic treatment of the PWYB led to the increase in the absolute number of B2- and the amount of B-lymphocytes, the absolute number of CD4+CD127+CD25- and T-activated lymphocytes, a decrease in the relative number of B1-lymphocytes and lack of fall of the level of T-regulatory cells, which have a positive impact on the clinical course of posttraumatic period in patients of the 2nd group: in 14-th and 30-th day it has increased frequency of relief of inflammatory symptoms as in the group as a whole as per patient and per recovery by 1 month after injury.

THE FEATURES OF IMMUNOLOGICAL REACTIVITY OF WOMEN YAMALO-NENETS AUTONOMOUS DISTRICT

Abstract

Conducted the expedition in the town of Nadym in Yamalo-Nenets Autonomous district (April, 2015). We examined 30 women, aged 40-64 living in the city of Nadym in Yamalo-Nenets Autonomous district. Immunological studies conducted in the laboratory of physiology of immunocompetent cells of the Institute of physiology of natural adaptations, VICKIE RAS, Arkhangelsk. In the peripheral blood were determined: the concentration of the phenotypes of lymphocytes CD16+, CD25+, CD71+, CD95+, HLADR+, the total number of leukocytes. The surveyed women in the city of Nadym in Yamalo-Nenets Autonomous district finds a reduction in the reserve capacity of the immune homeostasis due to the increased cytotoxic activity (CD16+) and apoptosis (CD95+) 56,66±2,49 and of 36,66±2,00 %, respectively.

Abstract

The experimental model of the in vitro transformed phenotype of neutrophilic granulocytes (NG) equipped with receptors CD64, CD32, CD16, CD11b, was created under the influence of fMLPs. The features of the transformed phenotype of NG were described. The influence of the Hexapeptide arginyl-alpha-aspartyl-lysyl-poured-tyrosyl-arginine (HP) on untransformed and transformed phenotype of NG was investigated in vitro. It was shown that fMLP increases the number of NG, expressing the receptor CD64, increases the density of expression of the receptor CD11b, and moderately reduces the amount of CD32+ NG. HP does not influence on the number of NG, bearing receptors CD64, CD32, CD16, CD11b, and the level of their expression. At the same time with simultaneous incubation with fMLP NG and HP, are shown to mitigate the effects of fMLP: reduced the number of CD64+ NG, reduced the density of CD11b expression, decreases in the number of NG bearing a receptor CD32. Overall, the results demonstrate the classic immunomodulatory effect HP: on the one hand, the absence of any change in the studied receptors of NG under his influence and leveling the transformational changes of phenotypic characteristics of NG, which arose under the influence of fMLP.

Abstract

The article contains the results of the morphological investigations when subcutaneously transplanting two types of the elastin biomaterial: structurized and dispersed one (patent № 2440148). The experiments have been carried out on Wistar rats (n=59). The transplanted biomaterials were subjected to the stage-by-stage resorption with further formation of the connective tissue regenerate. When doing it, the cells of the macrophagal and fibroblastic differentiation come out as a unified morphofunctional system in which giant multinuclear cells are an indispensable element. Depending on the biomaterial structure there are singled out two histotopographic types of the replacement regeneration: frontal type for the structurized biomaterial and diffuse-locunar one for the dispersed biomaterial.

Abstract

Aim of the study - to investigate the effect of erythropoietin (EPO) on the absorption and NBT-re-ducing capacity of neutrophils isolated from the blood of preterm neonates (PN) in vitro. The study was performed on 18 infants: PN from premature birth (PB) (n=6); PN from early preterm birth (EPB) (n=6); PN from a very early preterm birth (VEPB) (n=6). The EPO used in concentrations of 30 IU/l; 15 IU/l; 7,5 IU/l; 3,75 IU/L. Evaluated the absorption capacity and oxygen-dependent metabolism of neutrophils isolated from umbilical cord blood. Have PN from EPB and VEPB, EPO in in vitro experimental conditions increases, and PN from PB - reduces the absorption capacity and NBT-reducing capacity of blood neutrophils. EPO effect on the absorption capacity of blood neutrophils in in vitro experimental conditions does not depend on gestational age and manifests itself in small doses (3,75 IU/l and 7.5 IU/l), and NBT-reducing capacity of blood neutrophils in large doses (15 IU/l and 30 IU/l).

THE EFFECT OF LOCAL APPLICATION OF ERYTHROPOIETIN ON DEATH INDICATORS OF LYMPHOCYTES IN THE BLOOD IN THE DYNAMICS OF EXPERIMENTAL THERMAL INJURY

Abstract

To investigate the effect of local application of erythropoietin (EPO) in the composition of transdermal film (TDF) on the death's indicators of lymphocytes in the blood in dynamics of experimental thermal trauma (TI). The experiment was made out on 90 white nonlinear rats that were divided into groups: 1 - intact rats; 2 - animals with the TI and an aseptic bandage (n=40), 3 - animals with the TI application of the TDF with EPO (n=40). Modeling TI IIIA degree and area of 3.5 % was carried out by immersing the interscapular region of the skin of the animal in clean water with a temperature of 98-99°C 12 s. In the preliminary studies was developed TDP on the basis of sodium carboxymethyl cellulose with EPO at the dose of 500 IU/g. The dressing was carried out daily. The stady was conducted for 3rd, 5th, 8th and 14th days. Death's indicators of lymphocytes was assessed by staining cells with fluorochrome conjugated with annexin V and 7-aminoantipyrine D in flow cytometer "Navios". It is established that at TI at 3, 5 and 8 day increase in the number of lymphocytes with the phenotype of the Annexin-5-FITC+/7-AAD- and Annexin-5-FITC+/7-AAD+, 5 and 8 day reduced the number of lymphocytes with the phenotype of the Annexin-5-FITC-/7-AAD-. In the application of TDF with the EPO at the TI for 5 and 14 days reduced the number of lymphocytes with the phenotype of the Annexin-5-FITC+/7-AAD-, 8 and 14 days, reduced the number of Annexin-5-FITC+/7-AAD+ cells at 5 and 14 days increased lymphocytes Annexin-5-FITC-/7-AAD-. Thus, in TT the application of TDP with EPO leads to a decrease in blood lymphocytes with signs of apoptosis and necrosis.

Abstract

Children of 6-17 years old with systemic lupus erithematosus (n=10), with systemic sclerosis (n=10), with juvenile idiopathic arthritis (n=47) and conditionally healthy controls of corresponding age (n=30) were examined. It was revealed by flow cytometry that after 24-hours incubation (37 °C, 5 % СО2) the spontaneous and anti-СD3-stimulated amount of СD38-positive T-lymphocytes, T-helpers and cytotoxic T-cells in children with rheumatic diseases was decreased in comparison with healthy control.

Abstract

The levels of humoral immunological parameters in children of 2-17 years old with oligoarticular juvenile idiopathic arthritis (n = 98), with reactive arthritis (n = 26) and conditionally healthy children (n = 42) were examined. There were no differences in the concentration of C-reactive protein and antibodies to modified citrullinated vimentin between healthy children and patients with joint diseases. The level of antibodies to extractable nuclear antigens and the antinuclear factor titer in children with juvenile arthritis were higher, than in healthy children and patients with reactive arthritis. The frequency of antibodies to extractable nuclear antigens and modified citrullinated vimentin in children with juvenile arthritis and reactive arthritis was low, most often an antinuclear factor in these patients was detected.

Abstract

The clinical laboratory efficacy of the drug "Timogen®" (glutamyl-tryptophan) in the treatment of hospital pneumonia in patients with weight deficit was evaluated. Eighty-four patients with hospital pneumonia and objective signs of weight deficit were examined. Patients were randomized into two groups: 40 people received standard etiopathogenetic therapy and 44 patients used "Timogen®" metered dose nasal spray as a supplement to standard therapy. The drug was administered in the acute period of the disease in a daily dose of 100 mcg a 7-day course. The application of the "Timogen®" was accompanied by statistically and clinically significant reduction in the duration of the main clinical signs of the underlying disease, normalization of structural and functional parameters of the immune system and restoration of nutritional status.

Abstract

The efficacy and safety of using synthetic dipeptide alpha-glutamyl-tryptophan in the complex therapy of severe mechanical polytrauma of various nature were investigated. The trial included 327 people. Patients were randomly assigned to two groups: experimental and control (160 and 167 people, respectively). Both groups received standard combination therapy, but the experimental group received additionally the immunotropic drug "Timogen®" (alpha-glutamyl-tryptophan). The drug was administered intramuscularly in a daily dose of 100 mcg during 10-day course not later than 24 hours after traumatic injuries. The structure, the severity and the progress of the visceral pathology, as well as dynamics of the immune system were evaluated in the comparative groups. "Timogen®" facilitates reduction of the incidence of visceral complications, and if they develop, it decreases duration of the main clinical manifestations normalizing the structural, quantitative and functional parameters of the immune system and accelerating regeneration of damaged tissues in patients with severe mechanical polytrauma.

THE LOCAL IMMUNE RESPONSE IN PATIENTS AT EARLY POST-SURGERY PERIOD AFTER DENTAL IMPLANTATION DEPENDING ON THE SCHEMES OF ANTIBACTERIAL TREATMENT

Abstract

Successful performing of dental implantation is one of the major aims of dentistry. Surgery induces violation of the integrity of the mucosa, activation of the mucosal immunity, reparation and regulation processes, that can be measured by levels of IgM, IgG, sIgA and also IL-φ, IL-4, IL-6, IL-10, IL-17A, IFN-γ, TNF-α on the local or systemic level. Some antibiotics and phytotherapy can be applied to prevent potential onset of postoperative complications. Differences in its efficiency and action on the mucosal immunity can be measured with comparative assay of the features of the local immunity in the saliva of patients before dental implantation and after applying the therapies. Application of antibiotics leads to decreasing activity of pathogenic microorganisms and reparative potential of the mucosa cells, that is manifested in statistically significant reduction of the IgG and increased IgM and sIgM levels, whereas phytotherapy enhances activity of the immune system due to stimulation of marked increase in the level of IL-4, IL-10, IFN-γ and TNF-α in the patients' saliva.

Abstract

On the model of experimental tuberculosis of guinea pigs to study the effect of amino acid arginine on the course of the tuberculosis process. Experiments were carried out on 75 guinea pigs and 25 rats (weighing 200-250 g). To reproduce experimental tuberculosis, a museum strain of mycobacteria was used at the Central Research Institute of Tuberculosis H37Rv, 1993. Groups of animals: 3 groups of guinea pigs of 25 pcs. (Healthy control, infected with ILW and infected with ILT, receiving arginine daily 10 mg / kg body weight of the animal for a month) and 2 groups of rats of 25 pieces (healthy control, infected with ILW and infected with MBT). Determined in the blood nitrites and nitrates, total lipid peroxides by ELISA Biomedica GRUPPE and R & D Systems. Results. In animals lacking resistance to tuberculosis infection, increased plasma levels of nitric oxide derivatives caused by the use of arginine is an unfavorable factor contributing to the progression of tuberculosis.

Abstract

The study of blood cell counts and analysis of their changes in wrestlers and boxers made it possible to reveal special features of the population and subpopulation spectrum of lymphocytes in wrestlers and boxers of different qualifications. The sportsmen of power sports with anaerobic metabolism (wrestlers and boxers a significant decrease in the general population of T-lymphocytes) occured due to a decrease in circulation of main subpopulations of T cells: T-helper and T-cytotoxic. There were also a decrease in number of cells with adhesion receptors (CD11b), the number of B-lymphocytes and cells bearing apop-tosis readiness markers (CD95), opposite to elevated cell count with lymphocytes early positive activation(CD25) markers. An increase in cell number with receptors for interleukin-2 (CD25) with parallel decrease in the number of hematopoietic cell precursors and the main subpopulations of lymphocytes (T, B lymphocytes) is considered to be a reflection of the negative impact of intense speed-strength anaerobic regimen on immune system, and possible may be resulted in immunodeficiency in these athletes. The immune status of athletes going in for acyclic sports is characterized by marked cytopenia of relative and absolute numbers of T-lymphocytes, both T-helper and T-cytotoxic lymphocytes, and also CD16 and CD56 positive cells, belonging to congenital immunity factors, and also B lymphocytes (CD20), compared to sportmen groups of skiers and swimmers, and that reflects a decrease in intensity of immunopoiesis in anaerobic type of energy metabolism, and considered to be a significant factor, affecting immune system quality reaction and T- and B-cell cooperation reaction.

FEATURES OF PARAMETERS OF IMMUNE SYSTEM DEPENDING ON THE SPORT

Abstract

Immunological study of peripheral blood cells with subsequent analysis of data obtained during examination of professional skiers and swimmers, that meet presumably aerobic specific physical activity conditions, comparing to data obtained from professional wrestlers and boxers, that meet presumably anaerobic specific physical activity conditions, shows some special features in peripheral blood cell count, namely, higher leucocyte count and eosinophil content in skiers and swimmers, comparing to wrestlers and boxers, and lower content of mature neutrophil, but higher content of early neutrophilic cell forms in wrestlers and boxers, accompanied by evident decrease in mature lymphocyte and monocyte content in skiers and swimmers, comparing to wrestlers and boxers. A significant decrease in absolute and relative content of T-lymphocytes and both T-helper and T-cytotoxic lymphoid subpopulations, as well as CD16 и CD56 -positive cells and CD20 B-lymphocytes in anaerobic sportsmen were observed, comparing to those in aerobic sportsmen, that was due to immunopoiesis decrease in anaerobic specific physical activity conditions. Cell activation markers investigation led us to conclusion of favorable signs of current adaptation processes in cyclic sportsmen, derived from higher absolute B-cell early precursors content and higher CD20 mature B-cell content observed, and was considered to be B-lymphopoiesis augmentation in aerobic sportsmen. The groups mentioned above also reveal a rise in absolute amount of peripheral blood cells, possessing cell-cell and endothelial cell -adhesion receptors, as well as CD95 receptor possessing immune cells, involved in FAS-mediated cell apoptosis, and also a rise in early CD25 and late HLA-DR- activation receptors-possessing cell subpopulations, and rise in CD34 leucocyte precursor cell subpopulations, that demonstrate high capacity of immune cells to cooperate with each other in multisystem cooperation, that leads to greater adaptation potentials of immune system in aerobic sportsmen. In groups of wrestlers and boxers a twofold levels of secreted IgA were observed, and was associated with a decrease in secondary immune response-associated IgG level.

Abstract

The clinical and laboratory efficacy and safety of combined use of the antiviral pharmaceutical product (Adamantine family) and the complex preparation for etiopathogenetic therapy of influenza and ARVI in children were examined in the prospective clinical trial. The trial involved 90 randomized into 2 groups children aged 2-6 years with verified diagnoses of influenza and ARVI. Patients of the first group received the drug "Citovir®-3", children of the second group received combined therapy ("Citovir®-3" + "Orvirem®"). Evaluation of the efficacy and safety of the investigational drugs was performed by means of comparative intergroup statistical analysis of generally accepted for such nosology clinical signs and laboratory findings, frequency and severity of adverse events. The analysis of the results revealed that the use of "Citovir®-3" as a monotherapy and in combination with "Orvirem®" contributes to the reduction of the main manifestations of infectious process without statistically significant intergroup differences. Monotherapy with "Citovir®-3" leaded to a more pronounced increase in the level of secretory immunoglobulin A in the entrance gate of infection (saliva) and α-interferon in the serum. During the clinical trial no adverse events and abnormal laboratory findings were found in the comparative groups. The efficacy and safety of combined use of "Citovir®-3" and "Orvirem®" allow us to recommend this combination for treatment of verified influenza A in children aged 2-6 years. For the treatment of ARVI of another etiology, the combined use of the investigational drugs has no advantage as compared to the monotherapy with "Citovir®-3".

Abstract

The comparative clinical efficacy and safety of drugs "Citovir®-3" powder for oral solution, "Citovir®-3" syrup and "Immunal®" oral solution in the treatment of influenza and ARVI in children were examined in the prospective clinical trial. Ninety children aged 2 to 6 years with influenza and acute respiratory viral infections of unknown etiology were included in the trial in accordance with inclusion criteria. All patients were randomized into 3 groups, each of which received one of the comparative drugs as a means of etiopathogenetic therapy. The investigational drugs were used in the age-specific dosage variances as outlined in patient information leaflet. The analysis of the obtained results showed that the efficacy of the investigational dosage forms of the "Citovir®-3" (powder and syrup) against influenza and other ARVI exceeded the efficacy of the comparative drug "Immunal®" in the speed of normalization of clinical and laboratory parameters. The "Citovir®-3" (powder and syrup) contributes to statistically significant and clinically meaningful reduction in the duration of the febrile period and some catarrhal symptoms in comparison with "Immunal®" in influenza and ARVI. "Citovir®-3" (powder and syrup) facilitates the raise of secretory IgA production in the nasopharynx. All the investigational drugs were characterized as well tolerated. No adverse events or adverse drug reactions were found during the trial. Given the clinical and laboratory efficacy and safety profile of "Citovir®-3" (powder and syrup) they can be recommended for their inclusion in the treatment of children with influenza and ARVI.

Abstract

Injection of allogeneic erythrocytes to mice leads to the development of anemia, the appearance of antibodies specific to RE, whose production coincides with the production of anti-erythrocyte autoantibodies and are characterized by progressive growth. The results of computer studies have been shown that the similarities between theoretical and experimental kinetics were achieved by applying a signal to the clone located as far from the autoreactive clone as possible in the investigated fragment of the immune network. Herewith, all clones of the immune network fragment demonstrate a progressive, continual growth of activity, that indicates a breakdown in the state of tolerance. Thus, the emergence of systemic chronic autoimmune processes may be the result of an immune response to antigens that activate lymphocytes, which specificity is similar but not identical to the specificity of lymphocytes recognizing autoantigens.

Abstract

In healthy people, it has been established the possibility of induction of early and late apoptosis of T-lym-phocytes proliferated in vitro under physiological conditions through the transfer of both the cellular and humoral components of an autologous apoptotic T cell culture. It was determined that under equal conditions of cultivation of lymphocytes: a high cell density and a depletion of the culture medium, the transfer of components of the cells stimulated by anti-CD3 antibodies regularly increased the sensitivity of T-lymphocytes to apoptosis. While transfer of components of the culture with the dexamethasone exerted a modulating effect.

The identified differentiated characteristics of the phagocytic activity and immunophenotype in Helicobacter pylori infection and association with fungi of the genus Candida. Mannoproteins circulating antigen of Candida albicans affects the immunophenotype of neutrophils, inducing the expression of proapoptotic and reducing the expression of adhesion receptors and cytolysis.

Habitual miscarriage of pregnancy (HMP) is one of the leading positions in female infertility. The analysis of T cell differentiation in peripheral blood in patients with HMP has been carried out. A significant increase in the absolute amount of CD45RA+ CD62L+ (Tn) of naive CD4+ and CD8+ lymphocytes was established with a simultaneous increase in the absolute amount of CD45RA-CD62L- (Tem) СD8+ effector cells in patients with HMP compared to the control group. The obtained data require further study and prove the role of the immune system during the gestational process.

The purpose of the study have developed a new method of preoperative prediction delayed the formation of a bone block in spinal osteosynthesis external fixation device. The study included 111 patients with vertebral fractures with external fixation osteosynthesis, of whom 16 people were slow fusion of the bone tissue. We investigated the immunophenotyping lymphocyte, functional and metabolic activity of neutrophils, the concentration of the major cytokines and antagonists of their receptors. Determined that preoperative lymphocyte 2,4x109/liter and more associated with slow bone consolidation in more than 85 % of the cases investigated.

Abstract

In the research we have analyzed of the cytokine profile of mixed saliva of patients with complicated fractures of the mandible. Studied the levels of proinflammatory and anti-inflammatory cytokines and their dynamics in the treatment process with the conventional schemes and additional treatment with recombinant IL-1β. Indicators of the level of cytokines (Il-1-β, IL-2, Il-4, Il-8, Il-10, and TNFα) in mixed saliva were determined before treatment and 10-12 days of therapy. The study identified the initial increase of proinflammatory cytokines IL-β and IL-8 in all patients, which may indicate the tension of the local immunity in response to trauma. Patients receiving additional immunotherapy, these parameters returned to normal, and level of TNFa was increased, suggesting a high adaptive capacity of the local level of the immune system at use of Betaleukin.

The research has been conducted on white laboratory rats and their posterity during various periods of post-natal ontogenesis. Toxic damage of a liver was modelled by introduction of tetrachloride carbon to mature females. Estimated absorbing ability of cages of a monocyte-derived sprout of marrow and monocytes of peripheral blood. In a monolayer of cages calculation of a fagotsitarny indicator and fagotsitarny index was made. In the course a research oppression of absorbing and Killing activity of cages of a monocyte-derived sprout of marrow and monocytes of peripheral blood of experimental animals is established.

The role of the autoimmune component in the posterity generative function disruption of female rats with chronic liver injury of various genesis was studied. Using immunohistochemical methods it was established that in experimental rats there are inhibition of proliferative and increased apoptotic activities of spermatogenic epithelium, an increase in the titer of antisperm antibodies and a change in the concentration of proinflammatory cytokines. It has been found that excessive contamination by the phenol in blood of the women of the observation group is associated with: an increase of the relative and absolute CD127- content in comparison to the norm and the control group parameters; a decrease of the absolute and relative CD95+ lymphocytes content relative to the norm and control group. Simultaneously, the C/T genotype of the FAS C14405T gene under conditions of increased phenol content in the blood of women having reproductive disorders significantly enhances the risk of an increase of the relative content of CD127- lymphocytes, which modifies the mechanisms of apoptosis that play a key role in physiological course of the gestational process.

Abstract

The antibiotiko produktivnost given on studying and an antibiotiko rezistentnost of pro-biotic strains of bacteria of the sort Bacillus, and studying of effect of combined use of antibiotics and probiotics are presented in article. During the conducted researches patrimonial resistance of bacteria of the sort Bacillus to a tseftazidim, an aztreonam, a colistin and specific stability of B. cereus and B. licheniformis 31 to penicillin, a tsefiksim, a tsefepim, B. subtilis 534 to Chloramfenicolum is established. At the same time the maximal antagonistic activity is registered concerning S. aureus. The most fissile antagonist is B. subtilis 534, and the least fissile B. cereus IP 5832. Studying of effect of combined use in the conditions of in vitro allows to judge the prospect of sharing of B. subtilis 534 and B. subtilis 3 with penicillin, B. licheniformis 31 and tsefiksimy in relation to S. aureus, B. licheniformis 31 with penicillin concerning S. aureus, B. subtilis 534 and B. cereus with tsefiksimy - S. aureus and S. enteritidis, respectively.

Abstract

Serum samples, taken from 12-17 aged children with allergic and concomitant gastrointestinal diseases have been analyzed by ELISA for the total IgE, IL-4, IL-10, IgG, IgA and sIgE to H.pylori. IgE-response to H.pylori prevailed over IgA и IgG Abs and was enhanced in groups of children with allergic diseases and chronic gastroduodenitis, particularly during the exacerbation of allergy. Among children with erosive and ulcerous gastritis and also in group of non-allergic patients with chronic gastroduodenitis, the levels and frequency of sIgE to H.pylori were significantly lower than in healthy controls. The secretion of sIgE to H.pylori is supposed as one of the protective mechanisms under the conditions of chronic mucosal inflammation, associated with favorable outcomes of gastrointestinal diseases.

Abstract

The levels of sIgA, CagA, IL-17, sIgE-to H.pylori and to CagA have been analyzed by ELISA in the samples of coprofiltrates, taken from children with allergy and concomitant gastrointestinal diseases. Anti-H. pylori and anti-CagA sIgE have been elevated in groups of patients with allergic diseases in combination with chronic gastroduodenitis and among children with erosive and ulcerous leisons, being the most significant among the last group. Non-allergic patients with chronic gastroduodenitis were characterized by the highest concentration of IL-17 and sIgA, which was associated with the confirmed H.pylori infection. Our data demonstrates that sIgE to H.pylori, together with sIgA, represent a part of protective mechanisms during the H.pylori -assosiated gastrointestinal pathology.

Lymphocyte apoptosis increase and H1 histone protein increase are supposed to be indicators preceding the destructive changes in immunopoietic organs.

In the course of the study, it was established that in patients with chronic recurrent herpes viral infection, the parameters of the neutrophilic granulocyte test are increased, the phagocytic activity decreases, the serum level of interleukin-2 decreases, and the levels of all categories of lipid peroxidation products in the blood serum increase. The detected immunological and biochemical disorders were successfully corrected by a complex of therapeutic measures using the intravascular laser irradiation method

Abstract

Investigation of CD39 and CD73 expression on interstitial cells of calcified aortic valves of 31 patients was performed. Not significant increase (p=0,45) of CD39-CD73+ in cells from tricuspid aortic valves (TAV) comparing with bicuspid valves (BAV) was shown. Only from female patient's valves the difference became significant (BAV - 23,67 (19,43;29,80)%; TAV - 59,34 (49,17;69,55)% (p=0,002)). These data proves the hypotheses of purinergic signaling involvement in calcification suggesting that it is sex related process.

Abstract

The aim of the work was to describe the nature of various orofascial manifestations of primary immunodeficiency in patients in the register of the Sverdlovsk Region in comparison with data from literature sources. As a result of the study, it was possible to trace the significant correspondence of the detected pathological manifestations with identical PID groups. The most common signs can be called retention of baby teeth, candidiasis of the oral mucosa, angular cheilitis, multiple caries in combination with manifestations of primary immunodeficiency in other organs and systems.

Abstract

The issues of the effect of iron nanoparticles on biochemical blood indices and nonspecific immunity parameters were studied. It was demonstrated that the introduction of iron nanoparticles into the diet of animals did not exert its own influence on the biochemical parameters of the blood. However, it negatively affects the body of the bird being studied, as evidenced by an increase in these groups of concentrations of alkaline phosphatase and aminotransferases. The study of the dynamics of the leukocyte profile revealed only insignificant fluctuations of the studied parameters within the limits of the physiological norm.

Abstract

The study of the indices of systemic immunity in women with endometriosis, depending on the presence or absence of pathogens of genital infection. Pathogens of genital infection were found in 50 % of women with endometriosis in endocervix, endometrium, peritoneal fluid and in the foci of endometriosis. The most commonly identified were Ureaplasma spp, HPV and CMV. Changes in the indices of systemic immunity in women with endometriosis in comparison with the control group were more pronounced in the presence of pathogens of genital infection.

Abstract

The article presents the results of comparative analysis of cytokinemia (IL-6, IL-8, IL-10, TNFα) in various autoimmune diseases: autoimmune thyroiditis, valvular heart disease, psoriatic arthritis, rheumatoid arthritis, reactive arthritis, ankylosing spondylitis, chronic glomerulonephritis with end-stage renal disease, systemic lupus erythematosus. Severity of cytokine response was maximal in systemic lupus erythematosus and chronic glomerulonephritis. The greatest deviations from normal values in these autoimmune diseases were observed in TNFα and IL-8 levels.

Abstract

The effect of human chorionic gonadotropin (HCG) on apoptosis and production of IL-10 and TGF-β1 by isolated pools of natural T-regulatory lymphocytes (nTreg, CD4+CD25bright) and precursors of adaptive Treg (aTreg, CD4+CD25-) in the in vitro system was studied. As a result, we demonstrated that HCG induces apoptosis of nTreg and interferes with aTreg apoptosis, without affecting the production of IL-10 and TGF-β1 by these cells.

Abstract

The issues of populational immunogenetics are described in this article. In particular, the comparative characteristic of the frequencies of HLA II class genes and haplotypes in Nagaibaks population with the main populations of the Chelyabinsk region (Russian, Tatars and Bashkirs) is analyzed. Genetic distances on HLA II class genes between Nagaibaks population and some populations of Eurasia are constructed. The results of our research show, that population Nagaibaks is largely different from the population of Bashkirs of Chelyabinsk region on the frequencies of genes and haplotypes of HLA II class. According to the assessment of genetic distances on the HLA II class genes populations of Russian, Tatar population of the Chelyabinsk region and the Chuvash are the nearest to Nagaibaks population.

INFLUENCE OF OXIDATIVE STRESS ON POSTOPERATIVE PERIOD OF DIABETIC CATARACT SURGERY

Abstract

The purpose of this study is to reveal the state of the body's antioxidant systems using Western-blot enzyme immunoassay in tears for peroxedoxin-6 (PRDX6) proteins and / or fragments of its disintegration as a marker of protection against oxidative stress in patients with age and diabetic cataracts. Patients and methods. 136 patients (136 eyes) with cataract of various genesis were examined. The control group was represented by 25 volunteers who did not have eye diseases (50 eyes). Two groups of patients were examined. The first group included 78 patients with age-related cataracts. The second group included 58 patients with diabetic complicated cataract. The material of the study was fluids of the tear fluid the of the anterior chamber of patients with cataracts. To determine the active peroxedoxin-6 (PRDX6) and / or its degradation fragments, a Western-blot enzyme-linked immunosorbent assay was used. The expression of enzymes of protection against oxidative stress in samples of tear fluid was studied and a comparative evaluation of the level of activity of lactic acid antioxidant enzymes in oxidative stress after cataract surgery was given. Results. In the postoperative period in patients with age-related cataracts, the increase in the level of PRDX6 is determined, which is confirmed by the absence of complications of FEC. With a complicated cataract, the PRDX6 content is 6 times lower than in patients with age-related cataracts. Conclusion. If there is an active peroxiredoxin 6 in the lacrimal fluid and / or fragments of its decay, age-related cataracts are diagnosed, and this corresponds to a high level of protection against oxidative stress. In such patients, the postoperative period proceeds favorably. In the absence of active peroxiredoxin-6 and / or fragments of its decay in the lacrimal fluid, the presence of cataract due to diabetes mellitus is diagnosed. In the study of anterocameral fluid, a marker of development of metabolic disturbances in complicated cataracts and a 45 kDa transporter of hydrophobic compounds of the family of Sec-14p-like proteins was revealed in patients of group 2. Cataracts are considered diabetic, and in the postoperative period, the risk of developing PEER increases. The use of the method makes it possible to increase the accuracy of the choice of cataract treatment tactics.

The analysis of interrelations between cytokines and immunological indices of cervical mucus with transient and persistent HPV infection was carried out. With transient HPV infection, the balance of pro- and anti-inflammatory cytokines is maintained. With persistent HPV infection for 6 months, activation of local protective reactions aimed at involving effector cells in the pathogen elimination process was noted. With persistent HPV infection for 12 months, there is a change in the direction of immunological regulation towards anti-inflammatory cytokines.

We detected rates of IL-6, -8, -10, TNF-α, and CRP in plasma of patients with sepsis and polytrauma at 1-2nd and 5-7th days after admission in ICU. All patients were divided into groups with/ without MODS development. A rise in mediator levels in all groups of patients were identified. The maximum of detected increasing was in patients with MODS. There was not a clear cytokines-etiological agent dependence.

Abstract

The aim of research was to confirm that induction and regulation of autoreactive lymphocytes in model of the experimental autoimmune encephalomyelitis (EAE) in rats is carried out indirectly through idiotip-antiidiotipic (IAI) interactions with participation of the lymphocytes producing antiidiotipic antibodies with rheumatoid factors (RF) properties. Methods. Rats were immunized by the main guinea pig myelin protein (MMPgp). The kinetics of antibodies and autoantibodies formation during the immune response was investigated. MMPgp antibodies and MMP rats autoantibodies were determined by ELISA. RF was determined during agglutination test of tannised loaded IgG erythrocytes. The presence of IAI interactions was studied by competitive ELISA method. Results. Existence of IAI interactions between MMPgp antibodies and RF on the one hand, and, between MMP rats antibodies and RF on another hand was demonstrated. Association between RF high level in a blood and lack of clinical signs of EAE was found in immunized animals. Conclusion. The mechanism of induction of MMP rats autoantibodies during MMPgp immunization is mediated by IAI interactions of that clones of lymphocytes, which producing antiidiotipic antibodies with RF properties. Antiidiotipic antibodies with RF properties is carry out a specific negative regulation of a humoral immune response against a heterological antigen inductor of EAE, MMPgp through IAI interactions.

CYTOKINE PROFILE OF MICE IMMUNIZED BY COMPLEX OF RECOMBINANT OPRF AND ATOX ANTIGENS OF PSEUDOMONAS AERUGINOSA

Abstract

Currently, infections caused by Pseudomonas aeruginosa are an important medical problem. Their treatment is extremely difficult because of the broad antibiotic resistance of the pathogen. Objective: to study the effect of immunization with a recombinant antigens of Pseudomonas aeruginosa on the concentration of cytokines in the serum of mice. Materials and methods. Mice were injected intraperitoneally with recombinant 25 μg OprF and 50 μg of toxoid. The cytokine responses of mice were monitored by using flow cytometry analysis at 2, 4, 6 and 24 hours after immunization («FlowCytomix Mouse Th1/Th2 10 plex»). Results. The dynamics of the cytokine levels in the sera of mice was revealed: after 4 hours IL-1α, IL-5; 8 hours later, Π,-1β, IL-2, IL-4, and Th17/Th21/Th22 cytokines-IL-17A and IL-22 and IL-21; 24 hours later, IL-6, IL-10, IL-12p70, TNF-a and IFN-γ. Conclusion. Candidate vaccine against Pseudomo-nas aeruginosa based on the recombinant proteins OprF and aTox activates both cellular and humoral immune system with induction of Th1, Th2 and Th17 responses.

Aim of this study is researching the biological and mechanical features of the material, made of ultra-high-molecular-weight-poly-ethylene(UHMWPE) Studing the mechanical features showed that the material prepared with addition of Al2O3 and with using mechanical activation has the highest mechanical features, wear resistance in particular, in comparison with UHMWPE samples without Al2O3 or prepared without mechanical activation. was chosen for. After 60 days orthotopical transplantation this material in rats we did not fined signs of inflammation, cellular infiltration, destruction of material or bone-cartilage defect. Sample was in position of implantation without any shifts. Acquired data shows the perspective-ness of UHMWPE as a material for creating bioimplants for cartilage defects replacement.
